PT-141 (Bremelanotide) — research reconstitution guide

PT-141, a synthetic analogue of the peptide alpha-MSH, is supplied by My UK Peptides as a lyophilised (freeze-dried) powder in sealed glass vials. Lyophilisation preserves the compound during storage and transit; reconstitution is required before it can be used in aqueous-phase laboratory work.

This guide provides a step-by-step reconstitution protocol for research purposes only.

Required materials

  • PT-141 vial (lyophilised powder)
  • Bacteriostatic water (preferred reconstitution solvent) or sterile water
  • 1 ml sterile insulin syringes or transfer needles
  • Alcohol swabs (isopropyl alcohol 70%)
  • Gloves and appropriate PPE

Before you begin

Ensure your work area is clean and prepared. Wash hands thoroughly and apply nitrile gloves. Allow both the PT-141 vial and the bacteriostatic water to reach room temperature if taken from cold storage — this prevents pressure differentials during needle insertion.

Step-by-step reconstitution

  1. Prepare the vials. Remove the flip-off caps from both the peptide vial and the bacteriostatic water. Wipe both rubber stoppers with an alcohol swab and allow to dry.
  2. Equalise vial pressure. Insert a needle briefly into the peptide vial to equalise any internal vacuum. You may notice air being drawn in or expelled — this is normal.
  3. Add bacteriostatic water. Draw up the desired volume of bacteriostatic water (see concentration table below). Insert the needle at a 45° angle and allow the liquid to run gently down the inside wall of the vial — do not inject directly onto the lyophilised puck, as this can degrade the peptide.
  4. Allow to dissolve. Gently roll the vial between your fingers. Do not shake vigorously. Allow 2–3 minutes for full dissolution. The solution should become clear.

Storage after reconstitution
