Mannitol is a sugar alcohol commonly used as a stabilising excipient in lyophilised peptide formulations. This guide explains its role and what it means for your research.

What is mannitol and why is it in your peptide vial?

Mannitol (D-mannitol, or mannite) is a naturally occurring sugar alcohol found in many plants and commonly used in pharmaceutical and research applications. In the context of lyophilised research peptides, mannitol serves primarily as a bulking agent and cryoprotectant — it protects the peptide during the freeze-drying process and produces a more visually consistent, structurally stable lyophilised cake or puck.

When you open a peptide vial and find a white, firm, well-formed disc rather than a loose, scattered powder, mannitol is often one reason why. Without a bulking agent, many peptides would produce only a thin, poorly defined powder after lyophilisation — one that can scatter or adhere unpredictably to the vial walls.

Cryoprotective function

During the freezing phase of lyophilisation, the formation of ice crystals can physically damage molecular structures. Mannitol, along with other cryoprotectants such as trehalose and sucrose, incorporates into the freezing matrix and reduces the disruptive effect of ice crystal formation on the peptide. The result is a lyophilised product with better preserved structural integrity.

Is mannitol present in all peptide formulations?

Not necessarily. Whether mannitol (or another excipient) is used depends on the manufacturer's formulation decision. Some suppliers offer excipient-free peptides (sometimes called "neat" peptides) for researchers who require control over all components in their experimental environment. Where excipients are present, they should be clearly disclosed on the CoA and product labelling.
